Batch Release made easy!
Ik ben heel blij dat de Batch RelEASY-applicatie in oktober vorig jaar met succes is gelanceerd. Deze innovatie verbetert de operationele efficiëntie en zorgt ervoor dat de Quality Standards worden nageleefd. Batch RelEASY is een van de eerste applicaties die GxP-rapporten live genereert op het Self-Service Statistical Reporting Platform.
Wat is Batch RelEASY?
Batch RelEASY is een digitale tool voor het maken van rapporten die het genereren van release packages automatiseert en eenvoudiger maakt voor Remicade, Stelara en Simponi (P)FB materiaal op onze Bio sites in Leiden en Cork. Het haalt data op uit onze bronsystemen, zoals SAP, MES, eLIMS, Trackwise en binnenkort Comet, en organiseert deze in rapporten die nodig zijn voor batch release. Het is echter geen transactiesysteem voor batch release. Het registreert geen beslissingen, activiteiten of data. De gegenereerde rapporten vragen om een 'handmatige review stap' op volledigheid, door een QA Officer, voordat een QP ze beoordeelt voor batch release.
De reis – Succes is niet altijd direct een feit, en onze reis was daarop geen uitzondering. In eerste instantie probeerden we Tableau te gebruiken om releaserapporten te genereren, maar dat bleek niet haalbaar. Dit leidde ertoe dat we andere opties onderzochten en uiteindelijk Posit.co kozen als het platform van onze keuze. Posit biedt een betere manier om op R & python gebaseerde oplossingen te implementeren en stelt developers in staat om de open-source tools te gebruiken die ze kennen en waar ze van houden, met het gecentraliseerde beheer, de beveiliging en de ondersteuning die ze nodig hebben.

In december 2022 kregen we groen licht van de wereldwijde Digital & Technology Council om Posit voor Batch RelEASY-ontwikkeling aan boord te nemen en te implementeren. Budgetgoedkeuring volgde in april 2023 en we begonnen met de bouw van Batch RelEASY. Het platform werd binnen J&J omgedoopt tot het Self-Service Statistical Reporting Platform (SSSR) en in januari 2024 werd het opgeleverd als een volledig GxP-compliant applicatie. In de zomerperiode hebben we de gebruikersacceptatietests voor Batch RelEASY uitgevoerd en de kwalificatie afgerond. Als eerste gebruiker van dit nieuwe platform werden we geconfronteerd met tal van uitdagingen.
Door gebruik te maken van agile methodologie en het software development life cycle (SDLC) framework werd Batch RelEASY op 29 oktober succesvol gelanceerd op de Bio sites in Leiden en Cork.
“Have you heard, we produced another Golden batch last week!” Ideally this is something you hear a lot in our manufacturing plant. It shows that we are continuously improving our performance, producing more product with the same resources and equipment, and meeting our site’s ‘Must Wins’. In 2024 the term Golden batch was first introduced at the Leiden site and is defined as: a batch produced on target, in the standard timeframe, without any workarounds, managerial interference or quality issues, and can serve as a reference point for further quality and efficiency improvements.
With this definition, the efforts to reintroduce standard work in the plant were kicked off. Achieving a golden batch meant that everything went smoothly: there were no (major) issues that resulted in downtime or delays, no NCs needed to be raised, and the output was according to plan. Furthermore, the batch started on time and operators could continue their work without having to multitask. In short: producing a golden batch meant that it was a good day!
Improving step by step
The last phrase of the definition focuses on the fact that we can learn from these golden batches. What did we do then that we normally don’t or that we haven't been able to replicate yet? Learning from past performance is essential to continuously improve, and we can incrementally without the need to change controls or get fancy new equipment that takes a year to implement. We’ve already done it so let’s make it the standard. To find historical golden batches and learn from them, we developed a tool to visualise the 3 main elements of the golden batch definition: Lead/cycle time (did we make the batch in the standard timeframe), yield (did we produce on target), and deviation/NC free (no quality issues).

Pilot site Leiden
Leiden was chosen as the pilot site to map and visualize the continuous perfusion process, starting with Stelara. After many months of mapping, data validation and making visuals that would be easy to interpret and useful for the business, the tool formally went live in February 2025. During the testing phase a diagnostic was already performed on the data, and opportunities were identified where improvements could be made for Stelara that would help improve mainly yield and lead time. The tool also confirmed our main known challenges with Stelara. At the time of writing, teams in both USP and DSP are implementing solutions to bring Stelara not only back to target but beyond. Fermenter performance improvements, DPC loading and processing yield optimizations, and DSP awareness on critical handlings will help us produce more Stelara more easily and efficiently.
Celebrate more Golden batches
Very soon we will do the same diagnostic exercise followed by improvements on our other products, so that we’ll be able celebrate more often the production of another golden batch!
